DMEM Powder: What You Add at Reconstitution and How a 100 L Pack Converts to Litres
A lab that goes through several litres of DMEM a week pays most of its medium budget for water, cold freight and fridge space. One pack of high-glucose DMEM powder makes 100 L and keeps for 48 months at 2 to 8 °C. The same formulation as a 500 mL liquid bottle keeps for 12 months. - Volume. One 100 L pack makes the volume of two hundred 500 mL bottles.
- Cost. The pack lists for about the price of twenty-two of those bottles, so the powder itself costs a little over a tenth of the same volume bought as liquid. That is the price of the powder only: bicarbonate, water, filters and staff time come on top.
- Same formulation. The powder and this bottle list the same glucose, glutamine and pyruvate levels, with phenol red and without HEPES, so moving from one to the other does not change the recipe your cells see.
- Shelf life. The powder carries 48 months against 12 months for the liquid, so a pack can sit in storage four times as long before it is opened.
- Cold chain. Both ship on blue ice and both need fridge space; the powder is not an ambient-storage item. The difference is bulk: one pack of powder against two hundred bottles of liquid.

What is in the pack, and what is not
The pack lists phenol red, 4.5 g/L D-glucose, 4 mM L-glutamine and 0.11 g/L sodium pyruvate. It is listed without sodium bicarbonate and without HEPES. Our DMEM reference page gives the standard bicarbonate addition as 3.7 g/L at reconstitution. Serum is not in any basal medium and is a separate purchase; the listing names 10% FBS as the usual serum level. Do not read the pH or osmolality of the dry-pack listing as the values of finished medium; ask us for the lot data and we tell you what is available for that lot.

A lab that needs phenol red-free, low-glucose or glucose-free DMEM buys liquid. A lab that needs HEPES can add it to medium made from the powder: HEPES is listed as a sterile 1 M solution in 100 mL and as a 1 kg powder that stores at 2 to 30 °C and ships at room temperature. For large volumes the bulk powder, titrated in-house, is substantially cheaper per mole than the sterile solution.
Check the name on the order: the powder family
Four basal powders come as 100 L packs with the same 48 month shelf life, the same 2 to 8 °C storage and the same blue ice shipping.

Three of the four carry the same list price and pack size, so a wrong line on a purchase order is easy to miss until a 100 L pack of the wrong medium arrives. Where a listing does not say whether bicarbonate is included, ask us before you plan the batch.
From pack to medium: the checklist
- Water. Use high-purity, low-endotoxin water; the reference page gives the resistivity to aim for.
- Dissolve. Make up the powder for the batch volume you planned and mix until clear.
- Add sodium bicarbonate. The pack ships without it; our DMEM reference page gives 3.7 g/L for the standard formulation.
- Adjust pH slightly below target. The pH rises during filtration, so set it a little low.
- Sterilise by filtration, not by autoclave. A supplier video on preparing sterile media covers when to sterile filter the complete medium, and a second video shows medium being prepared from powder the same way.
- Add serum and antibiotics as supplements. The same supplier video groups serum, amino acids and antibiotics as the supplements added to basal medium.
- Keep the hood routine tight. A commenter on a DMEM preparation video lists what the demonstration skipped: loading the cabinet, and wiping down the filter unit, the serum bottle and the antibiotics before they go in.
- Label an in-house expiry and store dark. Once glutamine is in solution, treat the medium as having a working life of about four weeks at 2 to 8 °C, protected from light, as our DMEM reference page states.
When powder pays, and when it does not
The 48 months belongs to the dry pack; the medium you make from it has a short working life. So the decision is about turnover: powder pays when the lab uses each batch within its working life and has the water system, filters and hands to make it. A lab that uses a few bottles a month is better served by the 500 mL liquid with its 12 month shelf life. A core facility that supplies several groups can make medium in batches and still hold the unopened powder for years.
Mistakes that cost money
- Treating the powder as ready medium. It is listed without sodium bicarbonate and must be dissolved, completed and filtered.
- Reading the pack price as the price of finished medium. The saving holds only if the lab already has what it takes to make it.
- Ordering 100 L for a low-volume lab. The liquid's 12 months suits slow use better than batches of made-up medium.
- Ordering the wrong powder. DMEM, RPMI 1640 and MEM powders share one pack size and one list price.
- Expecting HEPES or a phenol red-free medium from this pack. Neither is in this powder; both exist as liquid.
- Planning ambient storage. The powder is a 2 to 8 °C item and ships on blue ice.
