MSC Extracellular Vesicle Collection: Serum-Free Medium or Depleted Serum, and How to Compare the Bottles
An extracellular vesicle (EV) programme can lose months at one purchasing decision: the medium the cells sit in while the vesicles are collected. Serum brings its own vesicles into the sample, and one research group describes switching cells into exosome-free serum for the harvest so the product is not contaminated with bovine exosomes. The alternative is a serum-free collection medium; we list three, in 500 mL bottles. The two-phase pattern matters for the order. The same group grew cells to high density with 10% serum, washed them, and only then moved them into the collection condition. So a programme usually buys two media: one to grow the cells and one to collect in. Why programmes take the trouble: mesenchymal stem cells show promise in regenerative medicine but come with challenges, and new research indicates EVs could deliver on that promise with lower risk.

Read the table as a buyer:
- Same listed limits, different price. The three bottles list the same endotoxin, osmolality and pH, yet two of them list at four times the price of the first. The listed limits do not explain the difference, so do not assume the bottles are interchangeable or that the dearer one is better for your cells; ask us for the lot data and we tell you what is available for each.
- None is listed for MSC by name. One names suspension and adherent mammalian cells in general, and the other two name no cell type. Plan a test with your own cells before you commit a run.
- One freight class. All three ship on blue ice and store at 2 to 8 °C, so they travel with most other media.
- Twelve months. The shelf life is 12 months, so buy for the runs you have scheduled, not for the whole project.

All eight carry a 12 month shelf life. The xeno-free bottles are frozen items that ship on dry ice, while the others ship on blue ice, so an order that mixes them arrives in two temperature classes and the freight is part of the comparison. The xeno-free bottle lists at about four times the basal serum-free bottle and includes the serum substitute, which the basal bottles are listed without. An MSC dissociation solution without phenol red, in 500 mL, stores at 2 to 30 °C for 36 months and ships at room temperature. One caution on labels. A supplier of a defined MSC medium renamed an attachment product from xeno-free to serum-free after finding it did not meet its own xeno-free definition. "Serum-free", "xeno-free" and "animal origin-free" are different claims; ask any supplier which definition applies to each component.
How to choose: a selection list
- Decide what the serum question is for you. If the readout cannot tolerate bovine vesicles, ordinary serum in the collection phase is out.
- Split growth from collection. Choose a growth medium and a collection medium separately, as the two-phase pattern above shows.
- Start with the bottle that costs least to test. The entry collection medium lists at a quarter of the other two.
- Run the routes in parallel. Keep your current route and the serum-free route side by side for a few passages and compare cell health and the readout you care about.
- Count the isolation step in. Isolating EVs from conditioned medium has its own considerations and challenges, and the medium is the first input to that step.
- Plan volume early. Until recently exosome manufacturing was limited to small batches; one developer that put manufacturing first from the outset reached thousand-litre batches.
What to ask any supplier
A researcher working with a company on an MSC-EV treatment writes that, as basic researchers, they struggle to understand what information they need to get for a clinical path. The medium is one of the places where that information starts. Ask every supplier the same questions:
- Which cells is the medium listed for, and is MSC among them?
- What do the listed limits cover: endotoxin, osmolality, pH, and anything specific to vesicles?
- If serum is involved, how is each lot tested for depletion?
- What does "xeno-free" or "animal origin-free" mean for every component?
- What lot data is available, and will the same formulation be supplied at larger volume later?
- What are the storage class and shelf life, and how does the bottle ship?
Mistakes that cost a programme time
- Collecting in the wrong medium. With ordinary serum medium, the vesicles in the serum end up in the sample.
- Paying for depleted serum without testing a serum-free route. The price complaint is public, and a serum-free collection medium can be quoted alongside it for comparison.
- Comparing on bottle price alone. Freight class and a 12 month shelf life change the real cost.
- Treating equal listed limits as equal products. Two prices sit behind the same listed endotoxin, osmolality and pH.
- Leaving the documentation question for later. Researchers moving toward a clinical path report not knowing what information to collect.
- Planning only bench volumes. Exosome manufacturing was limited to small batches until recently; ask about larger volumes before you standardise on a bottle.
